Product Detail The proteasome is a multicatalytic protease complex characterized by its ability to cleave peptides at Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group under neutral or slightly alkaline pH conditions. The proteasome possesses ATP-dependent proteolytic activity, and this subunit is involved in antigen processing to generate class I-binding peptides. Replacing PSMB5 with PSMB8 can enhance the ability of the immunoproteasome to cleave model peptides after hydrophobic and basic residues. It is involved in the production of spliced peptides formed by the ligation of two separate proteasomal cleavage products that are not adjacent in the parent protein. It serves as a major component of interferon-γ-induced sensitivity. It plays a key role in apoptosis through the degradation of the apoptosis inhibitor MCL1. It may be involved in inflammatory response pathways. In cancer cells, the replacement of isoform 1 (E2) with isoform 2 (E1) leads to a deficiency of the immunoproteasome. It is essential for the differentiation of preadipocytes into adipocytes.
