Components of the cleavage factor Im (CFIm) complex, acting as activators in the process of pre-mRNA 3' end cleavage and polyadenylation, are required for the maturation of pre-mRNA into functional mRNAs. CFIm helps recruit multi-protein complexes at specific sequences in the 3' end of pre-mRNA, known as cleavage and polyadenylation signals (pA signals). Most pre-mRNAs contain multiple pA signals, leading to alternative cleavage and polyadenylation (APA) that generates mRNAs with variable 3'-end formations. The CFIm complex serves as a key regulator in the selection of cleavage and polyadenylation sites during the APA process by binding to the 5'-UGUA-3' element in the 3'-untranslated region (UTR) of numerous pre-mRNAs. NUDT21/CPSF5 indirectly activates the mRNA 3'-processing machinery by recruiting CPSF6 and/or CPSF7. Binding to the 5'-UGUA-3' element located upstream of the pA signal acts as an enhancer for pre-mRNA 3' end processing.
