Propidium Iodide (PI) is a fluorescent dye that stains DNA within cell nuclei, commonly used for apoptosis detection. It is an analog of bromoethidine that emits red fluorescence after incorporation into double-stranded DNA. Although PI cannot penetrate intact cell membranes, it crosses damaged membranes to stain nuclei. PI is frequently used in combination with fluorescent probes such as Calcein-AM or FDA to simultaneously stain both live and dead cells. The excitation and emission wavelengths for the PI-DNA complex are 535 nm and 615 nm, respectively.
