SRB sulfonyl chloride (also known as lissamine alkaline red red) is highly fluorescent, but it is unstable in water and reacts with fatty amines, especially at high pH. It is usually modified for proteins at low temperatures (e.g.,at 4 °C). SRB sulfonyl chloride reacts with amine-bearing compounds (amino acids, peptides, and proteins) to produce a strong red fluorescent conjugate that is highly stable and resistant to protease-catalyzed hydrolysis. Unlike succinimidyl esters, sulfonyl chloride reacts indifferently with aliphatic and aromatic amines.
