The fluorescent dye PI (propidium iodide) is a nuclear staining reagent that can stain DNA and is commonly used in the detection of apoptosis. It is an analog of ethidium bromide that releases red fluorescence when embedded in double-stranded DNA. Although PI does not cross viable cell membranes, it does cross broken cell membranes and stain nuclei. PI is often used with fluorescent probes such as Calcein-AM or FDA to stain both live and dead cells. The excitation and emission wavelengths of the PI-DNA complex were 535 nm and 615 nm, respectively.
