Details

Bone metabolism is a dynamic equilibrium process of bone resorption by osteoclasts and bone formation by osteoblasts. Osteoblasts are responsible for the synthesis, secretion, and mineralization of the bone matrix. They act as the controllers for the activation or termination of bone remodeling. They play a crucial role in the trauma repair of bone tissue, the growth and development of bone tissue, and the balance of bone mass, and are the main focus in the research of bone diseases such as osteoporosis and fractures. The differentiation of osteoblasts is the prerequisite and basis for their leading role in bone formation. During the process of osteoblast differentiation, their most fundamental biological characteristic is the synthesis, secretion, mineralization, and maturation of the bone matrix. Mineralized nodules are the markers of the differentiation and maturation of osteoblasts. Meanwhile, they are also the main morphological manifestation of osteoblasts performing their osteogenic function. Observing the mineralized nodules of osteoblasts is one of the common methods for studying the differentiation of osteoblasts.

Common staining methods for the mineralized nodules of osteoblasts include the tetracycline method and the Alizarin Red S method. This kit uses tetracycline solution to stain osteoblasts. Tetracycline can form fluorescent phosphate complexes with calcium salts in mineralized nodules, which can be observed under a fluorescence microscope.
