In the process of tissue sectioning, when some tissues contain bone or calcification, the tissues containing calcium should not be directly embedded in paraffin sections. This is because the density between calcium and paraffin is different and it is more difficult to cut out complete sections. It is best to fix the calcium-containing tissue before decalcification or both. In the process of decalcification, acids break the transverse chains between protein molecules in tissues and release hydrophilic groups that can bind with water molecules, causing tissue swelling. After tissue decalcification, the acid in the tissue should be immediately neutralized, so that the tissue can be rapidly deacidified and neutralized to ensure the quality of the section and the effect of staining.

The alkali treatment solution after decalcification is mainly composed of sodium sulfate, lithium sulfate, etc.and it is recommended to treat overnight.
