Fecal samples are the most common samples for laboratory diagnosis of parasitic infections which consists of three independent steps: direct smear method, concentration method, and permanent staining smear method. The direct smear method requires fresh feces and can detect active protozoan trophozoites, protozoan cysts, worm eggs, and larvae; The concentration method can improve the detection rate of protozoan cysts, coccidian cysts, microsporidian spores, worm eggs and larvae, and there are precipitation and flotation methods; Permanent staining smears are easier to identify intestinal protozoa. Among them, the permanent staining method can confirm suspicious substances found in wet slides and identify protozoa not found in wet slides. Other samples from the intestine, such as duodenal aspirates or drainage fluids, mucus obtained by intestinal capsule method, and samples obtained by sigmoidoscopy, can also be used for permanent staining to detect protozoa. Among multiple staining methods, the most commonly used is iron hematoxylin staining and Wheathley trichrome staining.

Wheathley trichrome staining solution was originally developed by Gomori and used for staining tissue sections and cell smears. In 1951, Wheathley improved Gomori's technique by adding fixation and dehydration processes, forming a simple and fast staining program for intestinal amoebas and flagellates. Chromotrope 2R can stain nuclear chromatin, chromatin, nucleosomes, parasite eggs and larvae, bacteria, and engulfed red blood cells in a reddish purple color; Light green or solid green dyes can stain the cytoplasm of cysts, trophs, and other cellular components in a blue-green color. This product is commonly used for detecting, identifying, and distinguishing intestinal protozoa from background materials in fecal samples. For scientific research only, not suitable for clinical diagnosis or other purposes.
