Under suitable reaction conditions, β-glucosidase (GUS) can hydrolyze X-Gluc into blue substance, which is insoluble in the indigo substance in the nuclear tissue of the transgenic cells, and the site or locus with GUS activity shows blue color or blue spots, which can be observed with the naked eye or microscope.

GUS Stain can be used for biochemical activity analysis, immunoassay and histochemical staining of tissues and cells, mostly used for GUS gene expression analysis of transgenic plants.
