Thioflavin T (ThT) is a cationic benzothiazole dye that permeates cells and can be used to visualize and quantify the presence or fibrosis of misfolded protein aggregates or amyloid proteins in vitro and in vivo (e.g. plaques composed of beta amyloid protein found in the brains of Alzheimer's disease patients; PrP fibrillar plaques found in the brains of CGD patients). ThT exhibits enhanced fluorescence upon binding to amyloid proteins in tissue slices, and can rapidly and specifically bind to antiparallel beta fibrils formed by synthetic beta amyloid proteins (1-42), but does not bind to monomers or oligomeric intermediates. The excitation spectrum of fibrous β - sheet bound dye species exhibits a characteristic 120nm red shift, which can be selectively excited at 450nm, resulting in a fluorescence signal at 482nm. ThT is a beta amyloid (1-42) fibril oligomer and fibril probe with high sensitivity and specificity. It can be used for in vitro detection and quantification of amyloid fibrils by measuring fluorescence levels using a fluorescence enzyme-linked immunosorbent assay (ELISA) reader.
