Oxygen free radicals act on the unsaturated fatty acids of lipids to generate lipid peroxides. The latter gradually decomposes into a series of complex compounds, including malondialdehyde (MDA). The level of lipid oxidation can be detected by measuring the MDA level.
Under acidic and high-temperature conditions, malondialdehyde (MDA) can condense with thiobarbituric acid (TBA) to form reddish-brown trimethine (3,5,5-trimethyloxazole - 2,4-dione), with a maximum absorption wavelength at 532 nm. After colorimetry, the MDA content in the sample can be estimated.
