 Mycoplasma mycoides is a group of infectious mycoplasmas, including six members: Mmc, MmmLC, Mcc, Mycoplasma leachii, MmmSC and Mccp . These members cause a variety of diseases in ruminants ranging from mild to severe and fatal.

 

Mycoplasma leachii was originally derived from cattle in 1963 Isolated from synovial fluid, it can cause arthritis, mastitis and calf pneumonia. It is a facultative anaerobic bacterium. It grows rapidly and can cause turbidity in liquid culture media or the formation of large omelette-shaped colonies on solid culture media. Sometimes the diameter of the colony can reach 2 mm. It can metabolize glucose but cannot utilize arginine and urea. It can be distinguished from other members of the Mycoplasma mycoides family in that it can metabolize trehalose but cannot metabolize mannose and glucosamine. According to the results of DNA hybridization experiments and protein sequence analysis, its genome is 85% homologous to MmmSC, and is classified between Mycoplasma mycoides and Mycoplasma capricolum.The intermediate position of the original body is different from the two in metabolic pathways, antigenic properties and associated diseases. During serological analysis, Mycoplasma leachii often cross-reacts with other members of the Mycoplasma mycoides family, causing great interference in the diagnosis of infectious pleuropneumonia in cattle and sheep.

 

Mycoplasma leachii can be cultured in vitro and serum Scientific methods are used for identification, but culture methods are more time-consuming and serological methods have more cross-reactions. The PCR method is used for identification, which has extremely high sensitivity and specificity, and the detection time is short, and the results can be obtained in just a few hours.


This kit selects a membrane lipoprotein gene P67 with a conserved sequence for PCR. Identification: The primer was verified by BLAST to specifically target Mycoplasma leachii and had no cross-reactivity with other biological genomes. When 15 Mycoplasma leachii strains and 7 closely related mycoplasmas were detected, only Mycoplasma leachii produced specific bands, and no other strains were amplified. It can be seen that this kit is suitable for the detection and identification of Mycoplasma leachii.

丝状支原体族（Mycoplasma Mycoides cluster）是一组具有传染性的支原体，包括六个成员： Mmc（Mycoplasma mycoides subsp. Capri.）、MmmLC (Mycoplasma mycoides subsp. mycoides Large Colony，现已被重新归类至Mmc)、Mcc (Mycoplasma capricolum subsp. capricolum)、Mycoplasma leachii (即Mycoplasma sp. bovine group 7, Mbg7)、MmmSC (Mycoplasma mycoides subsp. mycoides Small Colony)和Mccp（Mycoplasma capricolum subsp. capripneumoniae)。这些成员在反刍动物中引起从较温和到严重致死的多种疾病。

Mycoplasma leachii最初是在1963年从牛关节液中分离出来，可引起关节炎、乳腺炎和小牛肺炎，属于兼性厌氧菌；其生长迅速，可引起液体培养基浑浊，或在固体培养基上形成大的煎蛋状菌落，有时菌落直径可达2毫米，可代谢葡萄糖，不能利用精氨酸和尿素，可与其他丝状支原体族成员相区分的特点是其能代谢海藻糖但不能代谢甘露糖及葡糖胺。根据DNA杂交实验和蛋白序列分析结果，其基因组与MmmSC同源度为85%，分类上位于丝状支原体和山羊支原体的中间位置，在代谢途径、抗原特性和关联疾病上均与二者有所区别。在血清学分析时，Mycoplasma leachii常与丝状支原体族其他成员发生交叉反应，对牛和羊的传染性胸膜肺炎的诊断造成很大干扰。

Mycoplasma leachii可以用体外培养法和血清学方法进行鉴定，但培养法较为耗时，血清学方法交叉反应较多。使用PCR法进行鉴定，具有极高的灵敏度和特异性，且检测时间短，只需数小时即可获得结果。

本试剂盒选取了一个序列保守的膜脂蛋白基因P67进行PCR鉴定，引物经BLAST验证为特异性靶向Mycoplasma leachii，与其他生物基因组无交叉反应。检测15个Mycoplasma leachii菌株和7种亲缘接近的支原体，只有Mycoplasma leachii产生特异性条带，其他菌株均没有扩增。可见本试剂盒适用于Mycoplasma leachii的检测和鉴定。
